Hey there---- I'm looking to read a murder mystery manga with a female protagonist. Romance is a plus but not required. I'd prefer if the manga had a dark ambience.
Thanks in advance!

Higurashi no naku koro ni and Umineko no naku koro ni series have a bit of a murder mystery feel, although not quite. They also don't have a female protagonist exactly, the narrator tends to be a guy (but it varies).
Doubt and Judge are also murder mystery style, much more than the previous two in fact, but again, I don't think they quite fit the female protagonist criteria.
All four series have hints of romance (if I remember well), but it's not really explored.

Gosick, don't let the cover and the art style fool you.

Yuureitou - highly recommended, though there is both male and female lead and you will learn of the female lead only after a chapter or two
