I have finally decided reading/watching shoujo after being repeatedly annoyed by a friend 
I love a good laugh or anything with blood, death and betrayals, in other words supernatural, horror and psychological such as Shin Angyo Onshi, Elfen Lied and Death Note. I'm already thinking about starting Hell Girl and restarting D N Angel, so something like that will do. Basically, anything that will be interesting to a guy who has only read shounen/seinen will be greatly appreciated.
Last but not least, no smut, shotacon/lolicon and yaoi please (I can handle shonen-ai but I probably won't like it much). Gender bender are fine since they're hilarious

Oh wow... I feel bad for ya, finally subjected to the horrors that are shojo. Well I'ma shonen fan myself so I'll tell ya some shojos that I liked...
Basara - More of a romance but it's got all the betrayal etcetc ya want. Genderbender but not so much for comedy. Currently my favorite shojo...
Tamago no Kimi - Just for the lol. Pretty light hearted.
Tsuki no Shippo Once again, mainly a romance (but hey when ya get into shojo it's mainly romance >.> ) but it's got some adventure and a lot of comedy.
Hana Kimi - If you haven't heard of this, ya better go read it. More comedy then romance and little to no blood stuff (I don't read much of that heh)
Mars - Psychological, trauma - filled. I didn't like it cuz it didn't have all that much comedy but I think it's to your taste...
Tokyo Crazy Paradise and Skip Beat! - You'll have to get over the horrible art but they're both hilarious. The former is a gender bender/action one and the latter is just lol.
All of the one's I named are romance. That's whatcha get for reading shojo >.>

Aishiteruze Baby Is ok.. I guess. Better then most non-horror Shojo.
I second Skip Beat! and Tokyo Crazy Paradise. Both are more then Hilarious.
These are Horror Shojo.. Dunno if you'll like them or not.

DN.Angel should be a good start for someone coming from the shonen department!
I'm also surprised nobody suggested Vampire Knight yet,its a darker,yet not too romantic shojo.
And then theres still Fruits Basket its cute and lighthearted at the beginning,but becomes pretty dark in the later.
For more funny stuff i suggest
Jun'ai Tokkou Taichou! - A delinquent couple kicking ass!xD
Kaichou wa Maid-sama! -A kickass school president working parttime at a maidcafe
Otomen -God,this one is hillarious,seriously!xD
[m]Ouran Highschool hostclub[/m] -Yeah,i know,the naruto of shojo manga,but still funny as hell!

faster than a kiss - the mangaka is a guy~~
ai shiteru baby - heartwarming tale of a playboy playing nanny, see Akachan to Boku too if you like this. Prepare to shed a manly tear or two.
sailormoon - shoujo super sentai! No mecha tho.

Sounds like shoujo equals torture. I'm in the opposite situation. I've been reading shoujo for a few years but just recently broadened my shounen reading beyond bleach and naruto.
Unfortunately you won't find many shoujos with blood, death, and betrayal. (One of the reasons I started reading shounen.)
A lot of the series already recommended are good reads.
I would add Mana
and maybe Hot Blooded Woman
If you're looking for comical gender benders I would add
Boku to Kanojo no XXX and Click

Personally, I think Koisuru Nikudorei-chan is so bad I can't believe it's not seinen or something (no offense!)...

Wow, defiantly agree. Go for the manhwa Saver. Perfect for you I think!
Also, Arcana is really really good too.
Then there's Vampire Knight, which I'm also surprised no one has mentioned. That's in top category.
