50 Rules for Teenagers.

It's not overly shoujo, there's a practical air to it, ...

I suppose I'm looking for a thoughtful romance with comedy involved.
*bows* Thank you in advance.

Tokyo crazy paradise gets somewhat deep later(midway to end or so). Only read if u can stomach violence. Girl is one amazing specimen of bamf later on.
skip beat isn't all that romantic even now, but it has been very steadily building the entire story(very long time) and yyet still doesn't seem stale to me... great story
Hana to akuma one of the best romance it was touching and I cried, I cried like a 10yo girl! *throws rock out the window*........"nyeeaark".
no bra probably a bit different than what you usually read but the romance is very sweet(and dangerous to male sexual orientation), and it does have its fair share of comedy.

Strongly second Hana to Akuma

you could also try:
Kimi ni Todoke quite unusaul situation, still very funny
Nineteen, Twenty-One webcomic and not very long, pretty good though
Hachimitsu to Clover very much comedy, but in addition to romances very serious views on how to live life, very interesting read
Orange Marmalade very funny, but also serious issues like persecution because of not being human

perhabs:
Akagami no Shirayukihime if you also like fantasy stories not very much comedy though
