Well, I doubt it's what you're looking for, but Yotsubato! has a pretty unique MC girl. I can't think of any other older, unique girls who aren't badass/tough, sorry.

What about Holo from Spice and Wolf by HASEKURA Isuna? The novel has both a manga and an anime adaption, and the female lead is not a kick-ass character but on par with the male lead.
Another unconventional female lead is Urabe Mikoto from Mysterious Girlfriend X by UESHIBA Riichi. I've seldom seen someone this strange.

unique in what way? since most manga have slight variantions of the same things, whilst out of those, nearly all of them are also unique in some way
