Okay, so i'm looking for something nice and really funny and quirky and cute, but you get a feeling of something serious underneath all the fluff. It would be a bonus if it had nice artwork. Like I want something that will really make me laugh, but will also make you care about the characters so much.

I'd prefer if it was an anime, just because I feel more like watching something, but I don't mind if its manga too.

Something similar would but Ouran High School Host Club, just coz I love Haruhi.
Clannad.
Angel Beats! is pretty similar.
and Fruits Basket

- It doesn't have to be like them, their all I could think of at the moment.

- It doesn't have to have a harem, but I like it whens there's a few characters around.

- I want a big romance, or at least you feel like its really important. But I don't want it to suddenly happen. I'd like it to build up.

- And I'd prefer it if the male fell in love first. Its okay if it's the other way around, so long as the girl isn't really weak and annoying and not willing to do anything about it.

- The setting doesn't matter so much, and the romance doesn't have to be the central part of the story, so long as you feel there's always something there.

- And like I said, I want something serious going on with one, or more of the characters. Whether they try and hide it or what, but it has to be important, and not just pushed to the back of the story when it comes out.

If anyone has anything that is even reasonable similar to what I have described, I'd love to hear your suggestions, please and thank you! =)

If you don't mind manga-only, I vote Kanata Kara. It's probably my all-time favorite.

BUT. You did ask for anime, so enough with that...

"Allison and Lillia" is all about adventure. Very cute, but serious in that it considers the effects of a war. And it's a multi-generational anime, which I found unique.

"Spice and Wolf" is very classy, cool characters, and it's about economics! Okay, so I'm not selling it well, but I promise it's worth it.

"Saiunkoku Monogatari" is certainly fun. Ah, as far as seriousness goes... one of the themes is how hard Shuurei must work to make any changes because she's a woman.

"Free!" a new swimming anime. So there's only like two episodes out. I guess I'm recommending it because Kyoto Animation is known for their stellar character interactions, development, etc. And it's cute boys being friends with each other doing cute things. What's not to like?

Kokoro Connect fits perfectly. It's both very hilarious and very serious at the same time. I reccomend to watch the anime.
Another good story is Toradora!. But it's less serious. Or better said, it's just your standard amount of drama that you can find in most stuff (but honestly I've to say that I read/watch all your four mentioned works and they're pretty much standard too from my point of view, so I suppose it should be ok). Again I think the anime is quite a good watch even if not as excellent as the novel.
And for something that I find often overlooked, I say that if you can consider Gosick "funny" (I'd say more cute than funny due to Victorique's character), it's also a good anime that can enter in the "funny but serious" category. In all its simplicity, it got one of the best romance story of the last years, so it's another title I think you should try.
Lastly there is Mahoraba. It's funny and I think it's far more serious than what it appears. The anime is kinda average though, I think the manga is better but I leave the choice to you.
The aforementioned Spice and Wolf above is a masterpiece. I'd not have brought up by myself since there isn't much comedy imho, but I'll vouch for its awesomeness since it's already being mentioned. The anime is an extremely good adaptation. I can't think how they could have adapted it better, but still, the novel remains superior.
Oh, and if you're up for an anime movie, I extremely recommend Ookami Kodomo no Ame to Yuki movie.

For manga only I say Molester Man is good. It's funny and there is some seriousness (the stalking part). But sadly I don't think its artwork can be considered "nice". Still if you can bear it, the story is good.

And I don't like to criticize other people's opinion... but Claymore... really? Why not Berserk then? laugh
