No she wasn't. They were step siblings and she never spoke to him and was extremely cold towards him. Not because she hated him, but because she was somewhat androphobic(scared of men).

The manga starts off with the brother sleeping and dreaming about being touched on the face by a ghost. Hes freaked out by it because it happens every night. One day he lays down on the couch and closes his eyes to go to sleep. Then he gets the sensation of being touched on the face by the ghost again and opens his eyes to see his step sister. After calming down from being surprised that he was awake she explains that to get over her fear of men shes been touching and kissing him while hes asleep. So he suggests that he help her out and the H stuff starts from there.
