Why does...(spoilers)
..Alexander have a snake mark on his face? Did he have anything like that in real life?

I have never heard that the real Alexander had a mark on his face.
Here, the mangaka seems to have taken artistic license to make the snake a symbol of Alexander. This was probably inspired by the fact that Alexander's mother, Olympias, belonged to a snake-worshipping cult and was thought to have slept with snakes in her bed. Olympias exerted great control over her son and played a large role in Alexander's political life. Arguably, her influence was poisonous.
The snake motif is seen several times throughout the manga, beginning with the first page where Eumenes picks up a device that is shaped like a snake.

I think it's a reference to the fact that Olympias put on a legend that she slept with Zeus in the form of a giant snake and conceived Alexander after that.

That makes a little more sense, so Alexander could be the son of a god...and I know enough about mythology to know the disguises gods used...
