Definitely Cyanide and Happiness at http://www.explosm.net
That is great for a laugh.
Gone with the Blast wave is also one of my top comics.

Nice choices. I kind of stopped regular reading online comics after rpgworldcomic died, but I check out some from time to time, like Penny Arcade.

I love Looking for Group and Misfile. Looking for Group however owns a piece of my soul.

Richard usually takes the whole thing. How'd you get away with only a piece missing? Did you distract him with a baby?

I read some that are listed but I also read these that aren't:

http://redallover.pensandtales.com/
http://spacecoyote.com/comics/sat/ - on hold but worth it.
The rain falls on my face
The smell of earth fills the air
You are not beside me
And what a cloudless sky above me

I believe there was an orphanage nearby that took precedence over soul stealing.

Saturday Morning Breakfast Cereal: http://www.smbc-comics.com/ - REALLY funny.
Perry Bible Fellowship: http://pbfcomics.com/ - Funny, good artistic style.
Fanboys: http://fanboys-online.com/ - It's funny with a pinch of drama every now and then.
Dueling Analogs http://www.duelinganalogs.com/ - Video game references galore.
XKCD: http://xkcd.com/ - Just like it's tagline says. Some of the romance-oriented strips are clever (and romantic).

used to read tons of webcomics, but now I'm only reading a few.
errant story from the same guy who brought you exploitation now. The comic that did everything wrong.
misfile has been mentioned already
so has applegeeks and megatokyo
Shadow Girls is a new webcomic that people should read.
Jack needs a mention for being the only furry comic I read.
alien dice Don't know why I read this one.
Level manga is freaking cool.
The Lounge My list wouldn't be complete without a little girl on girl action.
9th Elsewhere is a genius of a comic. If only it would update more often.
I used to read Tsunami Channelbut it went on hiatus for awhile until recently that is. I haven't had a chance to catch up. if I remember right Akira is doing tsunami channel in english for practice so there some grammatical errors here and there.
Underpower started out as a comic, but has recently degenerated to an art dump of sorts. It's still worth checking out.
Demonology 101 is probably the best web comic ever. I always wonder why its never been published. It's not like any of her other work hasn't been done in paper.
I used to read Strange Candy but I was so far behind when I started reading it I never caught up to the current strips and now I'm even farther behind. It also goes by the name Okashina Okashi.
Two sides wide used to be a crudely drawn four panel strip. Damn funny. Not sure what's happening now.
FOG Club Adventure went on hiatus, then the author came back and finished it.
Mixed Myth is probably the funniest fantasy webcomic ever with the most sarcastic, drunk, talking horse ever. That comic finished but I have yet to finish it.
That's about all the comics I can think of off the top of my head that are still online which I used to read.
Sarcasm just doesn't work over the internet.

Mmm... My absolute favourite webcomic's got to be Gunnerkrigg Court. So very good especially if you like:
Strong cool-headed heroines
Steampunk-modern like worlds with magic vs science societies
Dark Harry Potter-esque atmosphere.
Other ones I really enjoy are:
Alternate <== hilarious school life+parody+crack with snarky main character.
The Perry Bible Fellowship <== more hilarious crack for cheering up, especially recommended for those who liked The Far Side comics.
All In Your Head and My One True Uke <== both by the same artist. Gorgeous art, and a hilarious parody of BL with crack.
(yeah, you can really tell I like my comedies... 🤣 )
For something else a little more series, I occasionally like to read
Khaos Komix <== some really touching and well written slice-of-life about gays, lesbians and transgendered people.

In order of preference:
Xylia
(I also like Talismen by this artist, Barb Jacobs, but it's been discontinued for various reasons.)
Inverloch
Phoenix Requiem
The Dreamer
Tales of Pylea
Demonology 101
Dominic Deegan
Sinfest (suggestive, but not perverted)
Real Life Comics
Megatoyko
Juathuur
Erfworld
The Draconia Chronicles
Haru-sari
and my husband won't forgive me if I don't add for him:
XKCD
If you've ever thought or said "Nice Guys finish last" and really meant it, then you should probably read this LJ post by DivaLion. It's incredibly insightful whether you're male or female.
