I don't believe a story like this exists because it's anticlimatic.

The closest you'll get is One Punch Man, or one of those stories where only one boss exists and then it's done.

Well, I think Assassination Classroom fits? The final boss shows up right from the getgo and it builds up to then eventful end game.

Okay.. What about not only at the beginning, but next villains are significantly weaker than one??

How about Dragon Ball Super?

Ones that I can think of:

• Hikaru no Go: If Sai counts.
• One Outs: If Tokuchi Toua counts.
• Shingeki no Kyojin: If the Colossal Titan / Armoured Titan count.
• One Piece: If Shanks counts. I thought at first that Shanks was just some random throwaway passerby, but it turns out he's actually really high up in the ranks of things? I've only read up to around chapter 200 or so though, so I'm not sure if this counts.
• Maoyuu Maou Yuusha - "Kono Watashi no Mono Tonare, Yuusha yo" "Kotowaru!": Chapter 1 opens off with the final boss fight. And then after that it develops into an economics-focused deconstruction of the Hero vs. Demon Lord world setting. It's great.
• Sekai Maou: Another Hero vs. Demon Lord type setting. Here, Demon Lord is way more powerful than the Hero, but she has no desire to take over the world or anything, so it's a 4-koma slice of life comedy type manga from her POV.

Doryoku Shisugita Sekai Saikyou no Butouka ha, Mahou Sekai wo Yoyuu de Ikinuku. - He kills the demon lord "Emperor of Darkness" in the first chapter... the same guy that barely got stopped(not killed) by the previous party of heroes.
I Became a Legend a Decade Later After Telling my Comrades to Leave Everything to Me and Retreat First - He fights an army of demons, other things and the "Majin ruler" for 10 years... in the first chapter.
