So I'm a total sucker for characters that are really good hiding their emotions or bad at expressing emotion. A lot of awkward, funny, cute, and heart-wrenching misunderstandings can start from a simple seeming lack of emotion.
A character being unable to express themselves, or perhaps a character determined to hide their emotions and doing a good job of it, characters that are just plain hard to read... I really like these themes. My favorite would probably be purposely hidden emotion.
If anyone has any good recommendations I would love to read them<3

Yaoi is rife with examples of the stoic seme who rarely shows his feelings. As for humorous misunderstandings because of it, I can't really think of anything right now, sorry.
For ukes, this one is I suppose obvious, Yashiro in Tadayoedo Shizumazu, Saredo Naki mo Sezu and Saezuru Tori wa Habatakanai. As an example, one person comments that "he smiles even when he's mad, so be careful". It's not exactly full of cute and funny moments, though.
For a het example, I think maybe Chiyou yo hana yo. As I recall, the main character has trouble reading her boss/boyfriend. In general YOSHIHARA Yuki's male characters keep a straight face even when making a joke. See Sheet no Sukima for a short example.

Rakuda Tsukai to Ouji no Yoru "Drowning Fish" (chapter 03)
I feel like I keep recommending this to people lately. Probably because I just read it haha, oh well. The story, though, is not one of heartfelt misunderstandings.
Also these:

****** HWSK probably fits your request the best. The uke does not purposefully hide his emotions but he's really bad at expressing them, which makes for warm encounters as well as decisive turning-points along the story.

And what the heck, I'll just say this: Sono Yubi Dake ga Shitte Iru (the seme) every one knows this, and the whole thing is pretty cliché, but I don't care, said it anyway 🤣
