Came to mind after a looking for homophobes thread. I thought it isn't polite to say this in a looking for thread so I can say it here.
I hate homophobes 🙂 You are already in yaoi, loosen your anus (...not by all means literally) and receive a pleasure 🙂
Anyway, first option for me.

First option for me as well. It doesn't matter if one is or isn't gay from the start, if they find each other and are compatible, then let the good times roll. 😀

How realistically they portray the homophobia in a story is what makes it worth the read or not, in my opinion.
However, in general, I don't care which they start with or whether they maintain their sexual orientation. As long as there's well-written chemistry, it's all good.
If you've ever thought or said "Nice Guys finish last" and really meant it, then you should probably read this LJ post by DivaLion. It's incredibly insightful whether you're male or female.
From a bumper sticker I like:

i prefer when one isn't gay and slowly falls in love with the gay guy..

I prefer that they are gay from the beginning. Actually such yaoi manga are not that common. At least I haven't read many. Then again, I don't read many yaoi titles.
I was reading Boku no Koe - not a bad manga. But that the guys who are originally straight suddenly find other guys attractive and sleep with them without hesitation really gets a bit on my nerves. Yet this author is usually like that. Take it or leave it. However, I do think that the situation where someone who's initially straight finds himself liking another person of the same sex and sruggles with it can also become very cliched if it's not well written.

I'm a believer in 'love has no gender' so I chose the first one. I absolutely hate it when one of them (seme or uke) is straight and flirts and/or cheats with women. That doesn't happen often, but I still hate it. Because I'm obsessed with Koisuru Boukun, I guess I like one gay and one a homophobe, but I haven't read enough of that type to say I love it. Both characters being gay from the start makes things easier, but even when the uke says he isn't gay, most of the time you can't tell that by their actions. >.<

I'm fine with any situation really, but I guess my most preferable one would be the chemistry. What I look for in a yaoi manga is a situation where guys who have feelings for each other but aren't quite aware of it yet show the slightest feelings of attraction towards each other. Thus, they eventually kiss or something that gives away an obvious confession of love.
This isn't a yaoi, but Seven Days is a good example of what I like to look for in a yaoi/shounen-ai manga.

I enjoyed the situation when one is gay at Solfege. The gay (uke) wasn't much into the other guy and it was really interesting to know 'how it will be'. But again, newbies must be too open-minded.
