300... comic -> movie -> manga
Aside from it not making any sense, why tell the same story over and over again?
That's a story that has been retold ad infinitum for centuries. There's a reason for it too. It's an amazing story. There was a movie in the sixties called The 300 Spartans that inspired Frank Miller to write the story.
Anyway, Finding Forrester would fit nicely. Or This is Spinal Tap

A lot of teen romantic comedies could easily make the translation into manga - in essence, the plots and situations aren't really not all that different.
Things like:
Better Off Dead
The Girl Next Door
Say Anything
Can't Buy Me Love
etc.
If you think about it, these are all very similar to the a lot of the romantic comedy manga that's already out there.
nah...not a big fan of movie books.

I'm going ahead and replacing "American" with "English/Western," and nominating the Mad Max series for adaptation. The format would just work.
A manga version of LotR would also be interesting if done well enough--has it already been done?

I didn't even think of the Mad Max series, it just might work.

One movie:
CLOVERFIELD!
Edit: It appears that there actually IS going to be a manga made after Cloverfield! I predicted it!

I think the Alien movies could make a good manga series.
Also Blade Runner would make a good one to.

Star Wars. Such a huge universe and intricate characters to explore.

Stars Wars has been done over and over and over and over in so many different media formats. I think Star Wars has something in every single media format (comics, books, dvds, VCDs, cartoons, toys, etc). If they made a Star Wars manga, then it'd hafta have nothing to do with any of the current Star Wars series. I'd either hafta be set WAYYY in the past or WAYY in the future. I don't wanna see Skywalker, Vader, Chewie or Solo references cropping up.

If they could re-tell the game KotoR through a manga, that just might be the most awesome thing in the world.

Hmm I'd love to see the 1927 film The General as a manga. It's my favourite movie of all time and I don't know how well they could adapt it, but I'd love to see an attempt.
Now these aren't American movies, so not 100 percent on topic but also the 1960 version of Village of the Damned. They did make a rather sad excuse for a ermake years later that just failed to convey the same feeling as the original, and the sequal from '63 was just bad in general.
There is also a French film called Le Roi de Cœur full of mad men that I would enjoy seeing a manga adaptation of.
