Kimi wa Petto << I hated josei like hell until I came across this masterpiece, it made me open-minded.
Honey and Clover by Chica Umino... and Nana by Ai Yazawa.

To second other people-
Kimi Wa Petto- though I didn't like the ending.
Nodame Cantabile
Forest of Gray City
Vampire Girl by Fujiwara Kaoru. Short. Supernatural. There are vampires. It's one of my favorites.
Nana- though it is dragging on now, Paradise Kiss (technically shojo), and Last Quarter by Yazawa Ai
Dolis by Kusumoto Maki
On the lighter and sillier side-
My Darling My Flesh by Yoshihara Yuki
For the Sake of the Rose
also, in terms of mangakas
Fujiwara Kaoru: Vampire Girl is really good. She has also written a bunch of short story collections which aren't quite as good, but can be interesting: Fetish, Paradise, A Story of Long Ago, and Withdrawal Love
Kusumoto Maki: Dolis, The Funeral Procession of K, Mock Turtle Soup. A lot of it is sort of dark, artsy stuff, I guess...
Nina, My Love by Ikuemi Ryou is good.
Some people like Honey and Clover... I wasn't a big fan. Also not a fan of Midnight Secretary, Gokusen, or Suppli, though to each her own... Suppli sort of struck me as a less charming, less successful version of Kimi Wa Petto, though really the two lead characters aren't that similar, I guess. By which I mean, comparatively speaking, Sumire totally kicks ass. Everything I've read by Saito Chiho, including Waltz wa Shiroi Dress de and Honoka ni Purple (shojo) tends to read like a bad romance novel.

Helter Skelter is a good one, imo.

A Delicious Relationship has now become my ultimate favorite josei. It drags in the beginning then it heats up like a delicious beef consomme.

For some reason I'm so obsessed with Silver (FUJITA Kazuko)! It's fantastic, in my opinion. Yeah, the girl was/is sorta weak but she really does try to become stronger.I'll probably like another Josei more at some point... but right now Silver wins for me.

I haven't read a considerable amount of josei manga, but I loved:
Kabocha to Mayonnaise
Paradise Kiss
K no Souretsu
Helter Skelter is okay and I even previously thought that the art was ridiculously bad, but I became accustomed to it.
And Hyakki Yakoushou is intriguing, and some series of IMA Ichiko. Too bad I'm not in the mood to read the things in this demographic right now. (Currently, hunting for old shoujo/obscure seinen stuff).

Are You Alice?
Dawn - Tsumetai Te
Di(e)ce
Karneval
I think that's all.
Also, out of curiosity...am I the only one who hates Midnight Secretary tried to read before, thought there was too much smut XD?
Are You Alice?-interesting read
Heptagon -This one is actually my real favorite josei manga out of the other mangas I've read so far. The others don't really measure up to this one..in my opinion.
Usagi Drop-very cute/sweet
these are my josei favorites.

I hear you say "Why?" Always "Why?" You see things; and you say "Why?" But I dream things that never were; and I say "Why not?"
Nodame Cantabile
Honey and Clover
Kimi Wa Petto
Itadakimasu
Gokusen
Those are the best Josei in my opinion. 🙂

Petshop of Horrors - My first josei, and still my favourite by far.
di(e)ce
Deep Love - Pao no Monogatari - The only work of fiction that has ever managed to make me cry my eyes out.
+C: Sword and Cornett
Hyakki Yakoushou
I would like to say Karneval, but I'm afraid its plot isn't all that great. It is adorably cute, though 🙂
I loved Usagi Drop when the kid was still...well, a kid. After the time-skip, I didn't find it very interesting. I've read too many stories about high-school girls living in foster homes.
K no Souretsu isn't exactly the best, but it's one of the most unique things I've ever read, so it still deserves an honourable mention.

Of course you're not the only one! Midnight Secretary really sucks, imo...Too much smutty sex, too little story. I find all romance josei to be very boring, actually.

Pet Shop of Horrors because it's Pet Shop of Horrors
Paradise Kiss simply because the characters are so interesting and complex. Then again, I haven't read a LOT of josei, so...
