Hey everyone I'm looking for a romance that has a unique plot. I dont care if it's shojo/josei or shonen/seinen.

Glados is a bitch. She made me kill the companion cube, the only friend I'll ever have. 😢

About a girl who, when little, was the test subject for her brothers' new fencing and martial arts moves. She's now grown up and is the gang leader at her school. However, she gets into an accident and now her soul is in the body of a beautiful girl in another school. Lots of comedy and action scenes, and romance.
Lee Han-Ah used to be bullied in 2nd grade for having a “wide and flat” butt by a boy in her primary school and has hated him ever since. Now, in high school she is the celebrated, legendary captain of Il-Hwa Girls High School and she is seeking out a pretty boy boyfriend. What happens when she meets the mellow and seemingly perfect Yoo-Min?
Everytime Makoto sees beauty she'll throw up. And the first people she meets at her new school are the two very beautiful boys. She's also forced to join their club, Beauty Research Club. Idk, it's unique but it wasn't an amazing, amazing read.

A unique main character in my view. I think she has OCD.
Too lazy to post links. I'll just list names, okay?
- Lovely Complex (tall girl versus short boy)
- Bloodhound
- Koukou Debut
- Hare and Guu (strange)
I'm not sure if Hare and Guu has a manga though
but I think I heard it was a funny anime. -shrug-

If a good romantic manga is what you're looking for, you have to read Red River. Trust me, I don't like shoujo usually, but this is one series that I couldn't let go. And don't let the old school artwork stop you, it's an all time classic. Romance is very central, but it's really about the evolution of the characters as they deal with changing circumstances. Since it's a Fantasy/Historical manga, you've got wars, betrayals, and politics mixed in as well.
Some other good shoujos include: Skip Beat, Tokyo Crazy Paradise, Aishiteruze Baby and Nodame Cantabile.
As for more shounen/seinen, you've got Bitter Virgin, D-ASH, Karin, Superior and School Rumble. Don't know what you might like better, but I tried naming some more varied titles.

Oresama Teacher - wow, I keep recommending this one everywhere. Anyhow, it's super weird, but it keeps you engaged.
Miriam - excellent, wonderful, awesome. Takes place in a Western cowboy setting, very unusual, and starts out with the female protagonist as a plucky eight-year-old girl, then skips to her at seventeen.
Basara - one of the most epic shoujo ever, spinning a rich, emotionally intense tale of love and war, but if you don't want anything heavy or drawn-out, this one's not for you.
Toshokan Sensou: Love & War - just beginning scanlation, but well worth reading, and is set in a future where libraries have to literally fight for the right of freedom of expression. Even better is the anime Toshokan Sensou, but I think it's a little less romance-centered, although the chemistry is still definitely there.

Well, it is labeled as a romance, and I guess it can be considered to be a romance. However, it is a very strange romance. It certainly has a unique plot, though.
There is nothing in the description of the manga about the romance, but there is a romance between two of the main characters of the manga that is quickly presented in the first volume (actually it is first shown of the very first page containing panels). It has remained a constant part of the plot so far.

Hoshi no Koe
Utopia of Homosexuality
Music of Marie
And Fujoshi Kanojo is certainly unique.
