I haven't read that many, but I really enjoyed:
The Devil's Trill
Kindan no Koi wo Shiyou (and it's sequels)
Itadakimasu

You should definitely check out this manga-ka:
ENJOUJI Maki
And my personal favorites are NANA, Paradise Kiss, Kimi Wa Petto and Private Prince.

OHMI Tomu's works
Nana was my number one but it's dragging.
Paradise Kiss was really good also.
I like Yazawa Ai's other works also but I don't think their josei.
Kimi kara no Resume was my first josei fourth manga so it holds a place in my faves.

I'm sure most of the best have been said already, but my personal favorite is Fujoshi Kanojo.
It portrays the fujoshi lifestyle realistically and charmingly without going moe-moe or fanservicey like Fujoshi Rumi. Plus, the art is solidly good.

Quote from tsuto
Nodame Cantabile - There are quite a lof of good josei around, but I'm a Nodame fan!
Yeah, I agree with this too.
I just started reading josei lately and the other great one I've read are Shuukatsu, Itadakimasu, Chouyo Hana Yo and Private Prince.

I'd have to go with Usagi Drop. Though, I do like Gokusen--to be honest I wasn't aware it was a josei 'til I checked through my lists--it's not as good as UD.
Also, I think Cousin has some potential, but I'm still not too sure from what little's been released.

Yoshino Aki has some really sweet works like [m]kimi wa kirikira [/m]although only one chapter has been scanlated at the moment and the once shot non-chan to watashi and shuukatsu
and Enjouji Maki (private prince) is really good as well.

I'm an old fan of Kuramochi Fusako, but I have terrible memory, so I keep forgetting to buy her newer stuff, but one of the best Josei comics (and one of my all-time favorites) was something I read in the late '90s called Tennen Kokekko.

When I was younger, I was more interested in Shounen and Seinen. Mainly because I like guys...and Shounen and Seinen provide lots of guys for me. ^-^ I didn't like reading Shoujo because I thought girls make the biggest deal on stupid things. Like in most Shoujo, they're air-headed, they don't shut up, they're persistant, and they cry too damn much... That's why I don't read Shoujo...
But a couple years back, instead of reading Shoujo, I skipped and started reading Josei instead. And I have to admit, my favorite Josei manga of all time is Midnight Secretary. You know, vampires are overrated with that Twilight book and movie, but I kind of like this one.
