manga/anime related manga (i should probably say manwha) probably zippy ziggy, anime probably full metal panic fumoffu (dont really find many anime comedies funny), if its not anime/manga related the "Comedy of ALL Comedies" to me is Stepbrothers.

In terms of manga it would definitely be Gintama
Funniest manga I've ever read!
As for novels/books(that are in english), Terry Pratchett's Discworld series is the most hilarious. Especially the books with Rincewind

Beelzebub is the newest one thats pretty funny.
Of course Sexy Commando belongs in a whole different category of comedy.

Hmm, that's a toughie. I guess Angel Densetsu will always make me laugh. For anime, I think Kodocha and Jungle Guu are tied.
As a honorable mention, One Piece has a lot of priceless moments, but I wouldn't say it's a comedy of ALL comedies due to the multiple tragic backgrounds.

GTO was a hilarious read. Nothing memorable comes to mind from the anime I've watched.
Out of all forms of media I've come across though, Jackass 2 was easily the funniest thing I've ever seen. It had me laughing my ass off throughout the whole thing.

Quote from Sagaris
As far as anime goes, I'd say Sayonara Zetsubou Sensei is my preferred comedy series.
This. The art style is just so awesome too.
