I love Hotel..it's one of the best one-shot I've ever read. I even listened to Louis armstrong's song and follow the lyrics while reading the one-shot. 😛
As for me, the most depressing manga:
Princess and 7 seeds..in a very good kind of way.
The most depressing manga (in a bad and annoying) kind of way are Koizora and Life

saw somebody said ai-ren, i agree. i ended up crying some, which now that i think about i can't remember what the manga was really about. it's on my list of manga to re-read.
nana is also horribly depressing. actually, oh god, the only thing i can remember is
when nana (hachi i mean) found out she was pregnant and takumi offered to marry her. at this point i was squeeing because i was a complete takumi fan. then if i remember rightly he forced her to have anal with him. oh. dear.
i seem to remember crying at akuma de sourou. selected parts of red river were quite depressing, oh, and fushigi yugi: genbu kaiden. strangely, the angst does it more for me than any other of yuu watase's works (which were not depressing but stupidly overly-dramatic and annoying).
as for anime (i know it's off-topic but as others have said i've found anime more depressing)... the last episode of code geass r2 made me cry loads
i guess the way nunnally was screaming and the way he sacrificed himself for this "zero requiem" thing. i guess i felt guilty because i started to hate lelouch and thought he was an arrogant bastard towards the end of the series, then it was revealed he was doing it for a reason. i'm a sucker for stuff like that.
err and grave of the fireflies. and romeoxjuliet as well, not so much but still, with the music and everything... the manga was pretty butchered though.

Pieta was really sad, though it ended well.
Girlfriends (by Morinaga Milk) was getting really depressing by the end of the second book (i'm a sucker for unrequented love).
There were also some really sad moments in Berserk. Infact a whole lot of that is just depressing as hell.
I keep meaning to read Hotel but still havn't gotten around to it (cos my dam internet is buggy ><), though it sounds really good.

I don't read depressing stuff because, i already live enough depressing thing in my everyday life, when you feel depressed it's because the anime has been badly done... i have read a visual novel that was really sad but i didn't find it depressing just beautifuly sad... it's "planetarian", in an anime, i find the few last episode of shuffle to be quite sad but not depressing too...

I admit I did have a tear in my eye as I read Voices of a Distant Star.
Not with the anime though, I didn't like it very much >.>
The manga was a lot slower and did a better job of conveying that vast and growing distance between them... man-sniffle
Bradherley’s Coach was also depressing. I think it was because all that stuff happened to young cute girls that it was so sad... when they give up hope.. ;_;

I probably haven't read as much manga as everyone here but I recently read 2 depressing mangas:
Basilisk
This wasn't so depressing but...
since everyone dies...
and Tista
I found this more depressing and the ending actually made me cry... although it wasn't that sad... I really liked it though <~~ likes to read depressing things 😛

I love tragedy and I am not easily upset so this is a hard one.I thought Akuma no Ororon had a very tragic ending but X and Angel Sanctuary were quite depressing throughout. Count Cain was quite depressing as well. Diabolo would have been more depressing had it been a little longer but the ending was rather tragic. Knife is a good collection of tearjerking short stories. Death Note wasn't exactly uplifting either though I wouldn't call it depressing. However I was easily the most upset at the Code Geass ending. But I guess the fact it's an anime has a lot to do with that.
The pen is mightier than the sword...and considerably easier to write with.

Deep Love: Ayu no Monogatari
Although it was tragedy piled over tragedy over tragedy, I've got to admit I Q___Q'ed.
Also, as for a yaoi, Lovers and Souls really got to me.
