Hi! I'm looking for manga with a strong manly girl as the main character in which she's not making a fun of herself. Hope it makes sense. If the term transgender is discussed even better. No hentai. Thankssmile

Otomen - more like a third main character in this series, but she doesn't make fun of herself. (She can't even do that...)
He Dedicated to Roses - As much as I don't want to recommend a Hwang Mi Ri work, well... This features a girl dressed up as a guy in order to alleviate her stress.
Basara - Never got passed the first chapter, but I'm sure she does everything in her brother's name.
The Third - strong willed girl. I never read the manga. Only watched the anime version so I don't know how the manga (which is said to be the prequel) goes.

I've read Otomen and liked it although she's not the type of character I'm looking for. More like After School Nightmare but female variant. I like drama, adventure and such.
I only started reading Basara but I didn't like the art so stopped at the first chapter as well.
Oh, and I did browse through this category and others like that but I didn't find what I was looking for. They're always looked at in a comic way.
I'll give the rest recommendations a try. Thanks smile

Oh, in THAT case (genuinely masculine female characters w/ serious genderbending and w/out comedy that makes fun of it), consider these classics:

Orpheus no Mado
Oniisama E - BRILLIANT, but be prepared to scrape your crushed heart up from the floor!
Rose of Versailles
Princess Knight

More recent manga that might suit:

Hana no Kishi
Savage Garden
Saver
Operation Liberate Men - occasional comedic elements, but not the "make fun" sort
Hana no Asukagumi! - not trans per se, but a really tough girl with short hair

Claudine, definitely. A serious take on transgenderism by the queen of manly females, Riyoko Ikeda.
Hourou Musuko's secondary main character is a girl like this who might be transgender.
Himitsu no Hanazono (FUJII Mihona) - Like Orpheus no Mado, this girl disguises herself as a man in order to get an inheritance.
Fushigi no Kuni no Senichiya - This story is about a princess who is disguised as a boy to keep her father from killing her.
Paros no Ken - Another princess brought up as a prince. She falls in love with a laundry girl.
Revolutionary Girl Utena - A classic. The anime is even better.
Shitsurakuen - A fairly suckish knockoff of Utena.
