I haven't read the manga so I'm not sure how often it happens, but I know Sunako from Perfect Girl Evolution can kick butt when she wants to.

I think the lead from Tail of the Moon eventually gets to that point. (Haven't read it either - going by what I know of the story.)

EDIT: Ooh, almost forgot Faye from Cowboy Bebop.

Matsuri Special is a pretty damn awesome manga about a strong female wrestler (who is also a typical boy-struck teen).

Yawara! is about a girl who is a judo champion. She's really great, full of personality and also goes through rigorous physical training to maintain her strength.

Crimson Hero has a gifted volleyball player who keeps herself very fit.

And there's Claymore, of course - toughest women ever.

She does get to that point but not until very late. She is alright for most of the story but gets better near the end.

Toshokan Sensou: Love & War just one chapter out but there's the anime and it's not bad at all
